Abstract

Objective:To investigate the Effects of 1,25-(OH)2D3 on stimulation of osteoclast-like cells formation and bone resoption,further clarify the mechanisms of osteoclastic bone resorption regulated by 1,25-(OH)2D3. Method:Set up a model of bone resorption in vitro,to investigate the influence of different concentrations of 1,25-(OH)2D3 (0、10-10、10-8、10-6 mol/L)on the number of bone resorption pits .The effects of 1,25-(OH)2D3 on the expression of ODF in murine marrow cell was examined by in situ hybridzation technique. Result:1,25-(OH)2D3 could induce the formation of osteoclast-like cells and their bone resorption activity in a dose-dependent manner. The number and area of bone resorption pits and the expression of ODF mRNA increased with the increasing concentration of 1,25-(OH)2D3(0、10-10、10-8、10-6 mol/L). Conclusion:1,25-(OH)2D3 could modulate the bone resorption activity of osteoclasts and the expression of ODF mRNA in murine marrow cell in vitro,thus adjust the bone metabolism.
