Abstract

Objective: To study the influence on apoptosis and apoptosis pathway in human hepatoma SMMC-7721 cells by quercetin.Methods: SMMC-7721 cells were cultured in vitro,and MTT was performed to observe the proliferation.Flow cytometry was utilized for measure the apoptosis and cell cycle.The expressions of Caspase-3,Caspase-9 and Survivin in cultured supernatant were detected by ELISA assay.Results: Quercetin could inhibit the proliferation of SMMC-7721 cells in vitro at 10 to 320 μmol.L-1,which was concentration-and time-dependent.At a dosage between 40 and 160 μmol.L-1 in cultured cells,the apoptosis rates was elevated,and cell cycle was blocked in G2-M phase.Quercetin was able to increase the expressions of Caspase-3 and Caspase-9 proteins but decrease the expression of Survivin protein.Conclusion: Quercetin can inhibit the proliferation of SMMC-7721 cells via the apoptosis induction and cell cycle inhibition,moreover,Caspase pathway was involved in the apoptosis pathway.
