Transformation of Cold-induced Transcription Activator CBF1 into Cucumber(Cucumis satives L.)

Abstract

Taking the strong cold resistance cucumber‘Shandong No.5'as material,cold induced effect of transcription factor CBF1 gene on cold resistance of cucumber was studied.Cold-induced transcription activator CBF1(CRT/DRE binding factor)was amplified and cloned by PCR from the genomic DNA of cucumber‘Shandong No.5'and fused with CaMV 35 Spromotor and Nos terminator to construct plasmid pROK2-CBF1.CBF1 gene was introduced into cucumber by pollen tube pathway method.The transgenic nature of the regenerants was demonstrated by plant growth on medium containing kanamycin.The results showed that,PCR analysis confirmed that CBF1 gene was incorporated into the genome of cucumber,seedlings of transgenic plant soluble sugar content,water content was significantly higher than that of the control;the MDA content and the relative electrolyte leakage of cucumber leaves was significantly lower than that of the control,transgenic cucumber has a strong cold resistance.
