Abstract
Objective:To observe the Deduhonghua-7 pill's affection on the degree of CCl4 induced experimental hepatic and its effect on cytokines IL-1,IL-6,TNF-α,and to explore its mechanisms.Methods:The rats were randomly divided into 5 groups,except the normal control group,other groups were injected intraperitoneally with 30%(v/v) solution of CCl4 in olive oil to cause hepatic fibrosis;modelling the same time,the rats in Deduhonghua-7 pill high dose group,low dose group and colchicine positive control group were given intragastric administration Deduhonghua-7 pill with 0.62 g/kg and 1.04 g/kg and colchicine 0.4 mg/kg respectively each day.After the seven weeks,collected the serum and liver tissue,then perform the enzyme-linked immunosorbent assay(ELISA) to detect the liver indexes and serum cytokines TNF-α,IL-1,IL-6 levels and perform HE,Gomori,Masson staining to observe pathological changes of liver tissue.Use the Consensus of the diagnosis and treatment of liver fibrosis and Semi-quantitative assessment staging system Ishak of liver fibrosis established at Xi'an council 2000,to grading,staging and scoring the liver fibrosis.Results:Compared low dose group with model group,the serum and the content of liver tissue HA,LN,PCⅢ,ⅣC,Hyp,inflammatory activity and semi-quantitative,the content of serum cytokines TNF-α,IL-1,IL-6 reduced significantly(P0.05);the content of high dose group's serum HA and ⅣC,liver tissue LN and ⅣC,inflammatory activity and semi-quantitative,serum cytokine TNF-α reduced significantly(P0.05);content of positive control group's liver tissue HA increased,Ⅳ C decreased,had significant difference(P 0.05),inflammatory activity and semi-quantitative showed no significant difference(P0.05),content of serum cytokines TNF-α,IL-1,IL-6 increased,had significant difference(P0.05).Compared with normal control group,serum HA,PCⅢ,ⅣC,liver tissue LN,PCⅢ,ⅣC in Deduhonghua-7 pill low dose group and serum PCⅢ and ⅣC,liver tissueⅣC in Deduhonghua-7 pill high dose group showed no significant difference(P0.05).Conclusion:Deduhonghua-7 pill can significantly improve CCl4 induced experimental liver pathology,reduce hepatic indexes HA,LN,PCⅢ,ⅣC and cytokines IL-1,IL-6,TNF-α in serum,and have anti liver effect.
