Abstract
Objective: To establish a method of isolating the osteoblasts from newborn rat calvaria and use the AlamarBlue assay to evaluate the effect of fluoride on proliferation of rat osteoblasts. Methods:The enzymes digesting method was used to isolate the osteoblasts. The proliferative response to NaF was determined by the percents of reduced AlamarBlue. Results:The proliferation of osteoblasts was increased by NaF from 100μmol/L to 1mmol/L,whereas decreased by of and more than 2mmol/L NaF. Conclusion:Fluoride has a two-phase effect: low doses promote the proliferation of osteoblasts,high doses inhibit the proliferation.
