RAPD Analysis on Genetic Diversity of Four Populations of Plecoglossus altivelis from Zhejiang and Fujian Province

Abstract

Genetic diversity and genetic structure of four populations of Plecoglossus altivelis collected from Dongzhang Reservoir of Fujian province(DZ),Nanxijiang River of Zhejiang province(N8 and N9) and Qingjiang test field of Zhejiang Mariculture Research Institute(QJ),were analyzed by random amplified polymorphic DNA(RAPD).A total of 106 stable and distinct bands,including 47 polymorphic ones,were detected from the four populations by using 15 primers.The percentage of polymorphic locus was 44.34%.The results of data analysis on four populations showed that the value of percentage of polymorphic locus,Nei's gene diversity and Shannon's information index were 23.58%~35.85%,0.0868~0.1410 and 0.1285~0.2049 respectively.These results showed that their genetic diversity were low,we should strengthen the protection of the Plecoglossus altivelis resources.Clustering analysis by UPMG method showed N8 and N9 priority clustered together,while DZ and QJ cultured clustered in another branch.
