Postconditioning animal modeling of acute kidney ischemia reperfusion in rats

Abstract

AIM: To investigate the establishment of rat model of ischemic postconditioning based on the model of acute kidney ischemia reperfusion injury (IRI). METHODS: 40 SD rats were randomized into 5 groups: control group, ischemia reperfusion group (IR group) and 3 ischemic postconditioning groups (IPO1, IPO2, IPO3 groups). IR group was induced by clamping the bilateral renal arteries for 45 min. IPO1, IPO2, IPO3 groups were treated by different postconditioning methods after 45 min kidney ischemia. The postconditioning method of IPO3 group was sequential 20 sec reperfusion and another 20 sec ischemia for a 10 cycles. After 24 h reperfusion, the blood samples were draw from the inferior vena cava and the nephridial tissues were fixed. Then renal function was detected by biochemical methods, morphological changes and tubular injury score of nephridial tissues were examined by PAS staining under light microscope. RESULTS: The blood urea nitrogen level of IPO3 group was (27.9±3.2) mmol/L, the serum creatinine level was (232±49) μmol/L, and the tubular injury score was (382±48). Compared with the IR group, the above 3 indexes of the IPO3 group were decreased remarkably (P0.01), and the injury of kidney was reduced significantly. However, there was no significant difference between the IR group and the other 2 ischemic postconditioning groups (P0.05). CONCLUSION: The postconditioning method of the IPO3 group after the acute kidney ischemia can reduce acute kidney IRI and help us to establish the postconditioning model of the acute kidney IRI in rats.
