Abstract

RT-PCR, single-sided(anchored) PCR, RACE, step out RACE, DDRT-PCR, cDNA RDA and SSH are all the techniques of cDNA cloning based on PCR. The advantages and disadvantages of each technique were dicussed and compared with those of their counterparts. These techniques are now successfully used in the fields of isolation of numerous ESTs, construction of cDNA libraries, obtaining of differentially expressed new genes and cloning of cDNA in full length.
