Abstract

Objective:IL 15 was recently identified as a novel cytokine that shares many biologic properties with IL 2, our investigation was to study the effect of IL 15 on cord blood mononuclear cell (CBMNC) and peripheral blood mononuclear (PBMNC) .Method:CBMNC and PBMNC incubated with IL 15 at different concentration were cocultured with K562(NK sensitive) or HL 60 (NK resistant) at different ratio of effect/target. By using MTT assay to test the specific killing of cord blood and peripherl blood,using ELISA to test the interferon r (IFN r) and tumor necrosis factor a (TNF a) protein levels and using APAAP to assay the expression of CD 56 and CD 16 antigens.Result:①Anactivited citoxicity of fresh CBMNC was significantly lower than that of PBMNC.When incubated with IL 15,the level of CB NK cyoxicity was significantly increased and even could reach the level of PBMNC,the level of CB LAK citoxicity was significantly higher than PB LAK.②IL 15 also significantly induced IFN r and TNF a protein production.③When incubated with IL 15 theCD 56 antigen of CB NK was significantly increased witch suggest IL 15 might increase the cyoxicity of CBMNC by increasing the number of CD 56 NK cells in cord blood.Conclusion:IL 15 can significantly increase the cyoxicity of CBMNC.Our study suggests that IL 15 can be an alternative cytokine for the adoptive immunotherapy of malignant and nonmalignant disorders.
