Abstract

To construct a bait vector with the virus attachment protein VP37 of white spot syndrome virus,102 bases of it were truncated and a pair of primer to amplify the fragment by PCR were designed,and whether it is toxic to the yeast and its autonomous activation in yeast two-hybrid system was measured.The truncated gene of VP37 was obtained and inserted into PGBKT7 DNA-BD vector successfully.The sequence was measured correct.It was transformed into the yeast AH109 and was detected autonomous activity.The fusion proteins expressed by recombinant plasmids did not activate the expression of the yeast reporter genes: HIS3 and ADE2.But it activated MEL1.It had no toxicity to yeast strain.Other 2 reporter genes can be used in yeast two-hybrid system to screen the cDNA library and to trap the gene interacting with it.
