Abstract
Objective To explore the effect of N(2)-L-alanyl-L-glutamine on intestinal mucosa structure and cell proliferation in enterocutaneous fistula rats.Methods The enterocutaneous fistula rats were divided into total parental nutrition(TPN)groups(n=11)and TPN+glutamin(Gln)groups(n=13).The plasma glutamine levels were measured by spectrophotometry.The D-lactate and endotoxin levels were measured by modified enzymology spectrophotometric method and Limulum lysate method respectively.D-lactate and plasma endotoxin in vein blood were measured after 7 days of nutritional supports.The morphology of small intestine was observed by Haematoxylin Eosin(HE)staining.The proliferation index(PI)and S-phase fraction(SPF)of small intestine mucosa was obtained by flow cytometry.Results Compared with the TPN group,TPN+Gln group significantly increased mucosal villous height,mucosal thickness,crypt depth,and villous surface area.The plasma glutamine levels in the TPN+Gln group were significantly higher than in TPN group(P0.05).The D-lactate and endotoxin levels in the TPN+Gln group were significantly lower than in the TPN groups(P0.05).SPF and PI of the TPN+Gln group was significantly higher than in TPN group(P0.05).Conclusion TPN+Gln can improve the intestinal mucosa membrane structure and decrease the absorption of plasma endotoxin.
