Abstract

Objective To establish a method of constructing a living dermis equivalent in vitro. Methods The human fibroblasts were primarily cultured and the collagen of the rat tail was also prepared. Rat tail collagen、concentrated culture medium、sodium hydroxide、the fibroblast suspension in calf serum were mixed pro rata to shape collagen gel and form dermis equivalent. Results The collagen gel made fibroblasts have a suitable condition and the fibroblast collagen gel can be stretched slightly without being torn or deformed. Conclusion ①Using the rat tail collagen can form dermis equivalent, which was the key to provide the artificial skin. ②Based on the favorable biological traits of the fibroblasts in collagen gel, fibroblast collagen gel is a better model in the research of the fibroblasts interact with extracellular matrix and cells.
