Abstract
Objective To study whether caspase 3 is activated during apoptosis of bladder cancer cell induced by arsenic trioxide.Methods The cell growth were observed by MTT colorimetric assay and cell-growth curse.Flow cytometry was used to assay cell-DNA distribution.The expressions of caspase 3 were detected by immunocytochemical method and Relative activity of caspase 3 were studied by enzmye immunoasssay method and the activition of caspase 3 were tested by Western blot.Results The cell growth was significantly inhibited by the different concentration of arsenic trioxide.24 hour after the drug treatment,apoptosis rates of 4μmol/L and 8μmol/L arsenic trioxide groups are 7.58% and 10.18%,they have significant difference with control's(0.83)%(P0.05).In 4μmol/L and 8μmol/L arsenic trioxide groups,expression rates of caspase 3 are(36.37)% and 60.41%,they have significant difference with control's 8.01%(P0.05).In 4μmol/L and 8μmol/L arsenic trioxide groups,activaties of caspase 3 were 0.205 and 0.413,they had significant difference with control(0.073)(P0.01).Activations of caspase 3 were detected in arsenic trioxide-treated groups.Conclusion Caspase 3 is activated during apoptosis of bladder cancer cells induced by arsenic trioxide.
