Abstract
Objective To investigate the effect of sufentanil postconditioning on cardiac troponinⅠinduced by myocardial ischemia-reperfusion injury in rats and cardioprotection.Methods Male Sprague-Dawley rats [body weight 250(32 g)]were randomly divided into six groups(n =72):Sham(n =12):the coronary suture was passed,not tied,and maintained for 150 min;I/R(n =12):ischemia for 30 min followed by 120 min reperfusion;Pos(t n =12):three cycles of ischemic postconditioning(30 s of ischemia and 30s of reperfusion)at the onset of reperfusion following 30 min of ischemia;Sufen(LS,MS,HS,n =12 respectively):respectively administration of 0.75,3 and 15 μg.kg-1 sufentanil for 3 min at the onset of reperfusion following 30 min of ischemia.Heart rate and mean arterial pressures were measured at the following time points:end of stabilization prior to ischemia(baseline);30 min of ischemia;30,60,90,and 120 min of reperfusion;at the end of 120 min reperfusion,the rats were sacrificed for assessment of cTnI,infarct area,cell morphology of light and electron microscopy.Results No significant differences existed in heart rate between groups in all time points(P 0.05),mean arterial pressures were significantly decreased at 60,90,and 120 min following reperfusion(P 0.05)in I/R and at 90 and 120 min following reperfusion(P 0.05)in the LS group compared to Sham;cTnI and infarct area in Post,MS and HS group were significantly decreased compared to I/R group;in cell morphology of light and electron microscopy,the damage of myocardial structure was significantly reduced in Post,MS and HS group.Conclusion Sufentanil postconditioning could induce cTnI and protect myocardium against I/R injury.
