Anti-tumor and antioxidation action of 4-o-amino-phenol-4′-demethylepipodophyllotoxin ether

Abstract

Objective To investigate the antitumor and antioxidation action of 4-m-amino- phenol-4'-demethylepipodophyllotoxin ether(MDE).Methods Antitumor action was studied in vitro by MTT method,and in vivo by transplanted tumor of mice S180 and H22.Malondialdehyde generation in tissues of tats was measured with the thiobarbituric acid reaction.H_2O_2-induced red blood cell hemolysis was determined spectrophotometrically.Results MDE strongly in- hibited SGC-7901 cell proliferation at 48 h in a concentration-dependent manner with IC_(50) of 36.5(17.1~77.8) mg/L.In vivo experiments showe.d that MDE obviously inhibited the growth of S180 and H22,respectively.MDE also significantly inhibited spontaneous malondialdehyde generation or malondialdehyde generation induced by Fe~(2+)-ascorbic acid in homogenate of heart, liver and kidneys of rats,and hemolysis induced by H_2O_2.Conclusion MDE has anti-tumor and anti-oxidation action.
