Inhibition effect of c-myc antisense oligodeoxynucleotide on the proliferation and expression of c-myc gene in HL-60 cells

Abstract

Aim: To study the inhibition effect of c-myc antisense oligodeoxynucleotide(ASODN) on the expression of c-myc gene and the proliferation of HL-60 cells and to explore the possibility of ASODN as a drug for leukaemia gene therapy.Methods: After treated by c-myc ASODN,the expression of c-myc mRNA was detected by RT-PCR in HL-60 cells. The cell cycle was analyzed by flow cytometric analysis. The morphological changes were observed under invert microscope.The cell proliferation was investigated by growth curve and MTT method. Results: After sealing c-myc gene with ASODN,the expression of c-myc gene was inhibited. HL-60 cells in S phase decreased from 55.6% to 16.7%.The growth of cells was repressed and the ability of proliferation decreased.The c-myc ASODN acted as a specific growth inhibitor to HL-60 cell and the effect was dose-dependent and specific in sequence.Conclusion: The c-myc ASODN can inhibit the proliferation of HL-60 cells by inhibiting the expression of c-myc gene and may be a new gene drug to inhibit the proliferation of leukaemia cells.
