Expression of Interleukin-1β and Interleukin-1 receptor type I in colon tissue of chroic and comprehensive stress depression adult rats

Abstract

Objective To investigate the expression changes of Interleukin-1β and Interleukin-1 receptor type Ⅰ in colon tissue of chronic and comprehensive stress depression adult rats.Methods Twenty Wistar male rats were randomly divided into 2 groups: control group(n=10) and depression group(n=10).Except the control group,the depression group was given various kinds of stimulation for continuous 35 days.During the whole experiment,the body weight of the two groups was measured on the 0 and 35th day.By open-field test and fluid consumption test,the behaviors of the animals were tested.The expression of Interleukin-1β in colon tissue of adult rats was determined by RT-PCR,and Interleukin-1 receptor type Ⅰ was determined by immunohistochemical method.Results Compared with the control group,the body weight in depression group significantly reduced(P0.01) after various kinds of stimulation for continuous 35 days.After chronic stress,there were significant differences for total ambulation distance,average movement speed and total number of rearings between two groups in open-field tests(P0.05).There was no significant difference in sucrose solutions consumption between two groups before experiment(P0.05).There was a significantly reduced consumption of preference for sucrose solutions(P0.01) and an increased pure water consumption(P0.01) in depression group compared with control group after various kinds of stimulation for continuous 35 days.The expression of Interleukin-1β in colon tissue by RT-PCR method and Interleukin-1 receptor type Ⅰ by immunohistochemical method in depression group were both significantly higher than those in control group(P0.05),and there was statistic significance.Conclusion There are higher expression of Interleukin-1 β and Interleukin-1 receptor type Ⅰ in colon tissue of adult rats after chronic and comprehensive stress depression.Interleukin-1 β and Interleukin-1 receptor type Ⅰ may play important roles in the occurrence and development process of colon dynamic obstacle because of depression.
