Abstract
Objective To observe the effect of resveratrol on matrix metalloproteinase- 9( MMP- 9) expression in human small cell lung cancer cells,and then explore its possible molecular mechanism. Methods Human small cell lung cancer line NCI- H446 was cultured in vitro,and stimulated by different concentrations of resveratrol. The migratory and invasive abilities were assessed by chamber invasion assay. Expression of MMP- 9 and translocation of NF- κB p65 subunit were detected by Western blot. Transcription of MMP- 9 was detected by luciferase reporter gene assay. Results After the treatment of NCI- H446 cells with resveratrol( 50 nM) for 24 hours,the migratory and invasive abilities were significantly decreased( 37% and 33% inhibition, respectively). Western blot showed resveratrol could inhibit the translocation of p65. Furthermore,resveratrol inhibited promoter activity of MMP- 9 in a dose- dependent manner,and the promoter activities of NF- κB deletion mutants were significantly affected by resveratrol. Conclusions The inhibitory effects of resveratrol on NCI- H446 cells immigration and invasion are,in part,associated with the NF- kB- mediated MMP- 9 expression.
