Abstract

Objective: To establish blood brain barrier(BBB) model in vitro by way of cocultivation of human endothelial cells of vein(ECV304) and astrocytes of rat.This model should be easier repeated and more similar to the BBB in vivo,and the cell should have more degree of purity.Methods: There were three procedures:(1) to dissociate,pure and culture rat's astrocytes;(2)to establish blood brain barrier(BBB) model in vitro(endothelial cells/ astrocytes untouch cultured);(3)to assay model.Results:The BBB model in vitro established by way of cocultivation of ECV 304 and astrocytes was similar to internal blood brain barrier(BBB)in vitro,either in morphology,barrier function or in expression of endothelial cells specificity label.Conclusion:The BBB model in vitro can be established by way of cocultivation of ECV 304 and astrocytes.
