Reduced glutathione ameliorated the inflammatory pathology in rats with acute radiation-induced lung injury and inhibited the expression of IL-6 and TNF-α

Abstract

OBJECTIVE To explore the impact of reduced glutathione on the inflammation of the acute radiation-induced lung injury in rats.METHODS 40 female Wistar rats were randomly divided into control group,irradiation group,irradiation+glutathione low/high dose group,a single dose of 20 Gy were given to build the rat model of acute radiation-induced lung injury.HE slice was used to observe the pathological changes in lung tissue;cells from bronchoalveolar lavage fluid were counted and stained by Wright– Giemsa dye;RT-PCR was taken to examine the mRNA expression of IL-6 and TNF-αin lung tissue;ELISA used to measure the concentration of IL-6 and TNF-αin serum.RESULTS The reduced glutathione alleviated the lung inflammation in rat acute radiation-induced lung injury,decreased the number of inflammatory cells in bronchoalveolar lavage fluid,downregulated the mRNA expression of IL-6 and TNF-αin lung tissue,reduced the concentration of IL-6 and TNF-αin serum(P0.05).CONCLUSION Reduced glutathione can alleviate the inflammation reaction in rat acute radiation-induced lung injury.
