Abstract

Objective:To investigate the effect of diacetyldianhydrogalactitol(DADAG)on expression of apoptosis-related protein Bcl-2 and Bcl-X_L in mouse leukemia L1210 cells.Methods:The cytotoxic effect of DADAG on L1210 cells was determined using MTT assay.DADAG-induced apoptosis in L1210 cells was identified by flow cytometry and electron microscopy.The lev- els of Bcl-2 and Bcl-X_L proteins were examined by western blot analysis.Results:DADAG dose-dependently decreased the surviv- al rates of L1210 cells compared with control.Apoptotic peaks were detected by flow cytometry after 24-h treatment with DAD- AG at the concentrations of 12.0,17.2,24.5,35.0,and 50.0 mg/L.Treatment with DADAG 50.0 rag/L for 24 h induced shrinkage of cytoplasm and condensation and margination of chromatin.DADAG down-regulated the expression of Bcl-2 and Bcl- X_L proteins in a time-dependent manner.Conclusion:DADAG induces apoptosis of L1210 cells via inhibiting the expressions of Be]-2 and Bcl-X_L proteins.
