Abstract
Objective: To observe the inhibitive effect of baicalin on the growth of human leukaemia cell K562 and its effects on cell apoptosis ratio.Methods:K562 cells were in vitro cultured.The concentration of cells in the logarithmic phase was adjusted to 1×105/ml.Baicalin was added into every pore,and its final concentration was adjusted to 25.0,50.0,100.0,200.0 and 400.0 μg/ml,respectively.Each concentration was repeated by four times.Cells were cultured for 48 h and 72 h respectively.The cell apoptosis ratio and cell cycle were tested by MTT colorimetry.The concentration of K562 cells in the logarithmic phase was adjusted to 1×107/ml,and they were treated with baicalin at 50.0,100.0 and 200.0 μg/ml for 48 h.Cells in treatment groups and control groups were collected.The percentage of cell apoptosis and cell cycle was evaluated by flow cytometry and software calculation.Results: Baicalin can inhibit the growth of human leukaemia cell K562 in a dose-and time-dependent manner.The K562 cell apoptosis ratio induced by baicalin 50.0,100.0 and 200.0 μg/ml was 1.3%,2.0% and 12.1%,respectively.The obvious apoptosis peak(AP) was observed.Conclusion: Baicalein Could inhibit the proliferation of human leukaemia K562 cells,its mechanism might be related to promoting the apoptosis of K562 cells.
