Effects of cell cycle and its correlated protein in cervical cancer cells after ~(60)Co gamma ray fractionated irradiation

Abstract

Objective To observe the effects of 60Co gamma ray with different fractionated doses on the cell cycle,cyclin D1 and B1,cyclin dependent kinase (CDK)1and 4 of cervical cancer Hela cell line.Methods0,2,4,6 Gy 60Co gamma ray fractionated irradiated one time per day for 5 days.Flow cytometry was used to detect the cell cycle.The expression of cyclin D1, B1 and CDK 4,1 were detected by RT-PCR analysis.Results2,4,6 Gy of 60Co gamma ray fractionated radiation caused G0/G1 and G2/M phase cells and apoptosis rate increase gradually,compared to the control group(P0.05).Cyclin D1 and CDK1 were not expressed in all irradiated groups,while the cells of control group expressed normally compared to the control group,the expression of cyclin B1 and CDK4 were suppressed in all irradiated groups(P0.05). ConclusionsHela cells arrest in G0/G1 and G2/M phase after 2.0,4.0,6.0 Gy of 60Co gamma ray fractionated irradiation, and at the same time apoptosis occures. The expression levels of cyclin D1,CDK4,cyclin B1 and CDK1 are suppressed after irradiation.
