Abstract
To express the recombinant human FS315 C termin al peptide and to prepare its antibody, the prokaryotic expression plasmid pGEX-FS315C for FS315 C terminal g enes was constructed, and the anti-human FS315 C terminal polyclonal antibody was prepar ed by immunizing New Zealand rabbit with FS315C protein. The results showed that the r ecombinant pGEX-FS315C plasmid highly expressed GST-FS315C fusion protein in E.coli B L21, and anti-FS315C terminal polyclonal antibody was obtained by immunizing New Zealand rabbit with GST-FS315C fusion protein purified by affinity chromatography and electrophores is. The antibody against FS315 C terminal only specifically binds to FS315 by Western blot, but h as no binding with FS288. By ELISA, the antibody against FS315 C terminal has specific reaction wit h FS315, but has no cross-reaction with FS288, inhibin or activin. The immunohistochemistry show ed that FS was expressed in macrophages with the antibody to FS315 C terminal, which was consis tent with the previous report. The specific anti-FS315 C terminal polyclonal antibody was successfully prepared by immunizing New Zealand rabbit with recombinant GST- FS 315C.
