Abstract
Different numbers of embryonic stem cells were injected into the different-period embryos of Meriones unguiculatus to research the effects on the embryos development and investigate the suitable conditions for making chimeric embryos. The experiment results indicated: The numbers of microinjecting ES cells(2~5,6~10,11~15), had no significant effect on both the rate of embryos development and hatch; but when microinjected with 11~15 ES cells, the rates of embryo survival and hatch were both the lowest, being 82.14% and 64.29% respectively; when microinjected with 2~5 ES cells, the rates of embryo survival and hatch were both the highest, being 90.90% and 78.79%. When microinjected with 11~15 ES cells and no microinjection, the rates of embryo survival were 95.12% and 82.14%, the difference was extremely significant(P0.01); the rates of embryos hatch were 90.48% and 64.29%, the difference was also extremely significant(P0.01). The blatula with no microinjection, microinjected with 8~16-cells of stage-embryos and blastula,the rates of embryo survival were 94.92%,91.53% and 87.50%, the defference was not significant(P0.05); but with no microinjection blastula and microinjected with 8~16-cells of stage embryos, the rates of embryos hatch were 89.83% and 61.02%, the difference was extremely significant(P0.01).
