Abstract

Objective:To study the effect of allograft immune tolerance induction by donor IL-10 and methylprednisolone (Medron)modified dendritic cells(DG)on allogenetic skin transplantation in mice so as to offer theoretical and practical basis for anti-rejection of transplantation.Methods:The recipient BABL/c mice were divided into six groups.Apart from group A,every group was injected with corresponding donor DC through vena caudalis,and transplanted full-thickness skin through back to back from donor C57BL/6 three days later.Corresponding doses:group A was injected with NS,as blank control; group B,non-modi- fied donor DG; group C,20μg/L IL-10 group; group D,10mg/L Medron; group E,20μg/L IL-10+10mg/L Medron group.Group F was skin transplantation between two BABL/c mice.Skin transplantation was performed and the survival time of skin allografts were observed.Results:Statistical analysis showed that compared with the other groups,the graft of group E survived longer(P<0.05).Conclusion:The pretreatment with donor DC treated by IL-10 and Medron can prolong the survival time of mice skin al- lografts after transplantation and induct immune tolerance successfully.
