Effect of TCM Mixture on Expression of Nephrin and Podocin in Podocytes of Early Diabetic Nephropathy Rats

Abstract

Objective:To observe the effects of Traditional Chinese Medicine mixture(TCM mixture) on the expression of nephrin and podocin in podocytes of diabetic kidney disease(DKD) rats. Methods:Rats were fed a high glucose and high fat diet and given a low dose of STZ to develop rat model of type 2 diabetes. Then the successful induced models randomly Divided into 2groups: TCM mixture treatment group and model group. In addition,the normal rats served as a normal control group. All the rats were received daily gavage respectively for 8 weeks. The concentration of blood glucose and urinary microalbumin were monitored before treatment and on week 4,8and 12 after treatment. twelve weeks later,the body weight(BW),kidney weight(KW),KW/BW,blood urea nitrogen(BUN),serum creatinine(Scr),cholesterol(CH) and Triglyceride(TG) were detected by biochemical methods. The pathological changes of kidney were also detected with optic microscope. The expression of nephrin and podocin in podocyte were detected by immunohistochemistry. The protein levels of nephrin and podocin in the kidney tissue were also determined by western blotting. Results: after TCM mixture intervention,the levels of urinary microalbumin,renal index,blood glucose concentration,BUN and Scr in were significantly lower and body weight was higher than those in STZ group(P 0. 05). Meanwhile alleviating the damage of kidney. The expression of nephrin and podocin were improved(P 0. 05). Conclusion:The TCM mixture protect kidney against possible related to improving the expression of the nephrin and podocin.
