Abstract
Objective To investigate the effect of human telomerase reverse transcriptase(hTERT) gene on human tongue cancer cells. Methods hTERT gene was cloned by means of RT-PCR and reversely inverted into retrovirus vector pLNCX to construct antisense recombinant plasmid. Then the recombinant plasmid was transfected into Tca8113 cells using liposome. Combinded with conventional telomeric repeat amplification protocol(TRAP), DNA seqence analysis technique was used to assay telomerase activity. Results After Tca8113 cells were transfected with antisense plasmid , the telomerase activity of them significantly decreased, and the rare of growth and proliferation was also slowed down in comparison with that before transfected. Conclusion The expression of inverse hTERT gene can effectively suppress the telomerase activity and the growth of Tca8113 cells.
