Abstract
Objective To investigate the protection effect of hydroxypiperquin phosphate(HPQP) on hematological system injury induced by radiation damage in mice.Methods 40 male Kunming mice were randomly divided into five groups(low/middle/high dose groups,blank control group and normal group).All except the normal group underwent whole-body γ-ray irradiation at 4.5 Gy.The peripheral leukocyte and platelet counts were determined for the primary effect on hematological system to screen out the most suitable dose of the drug.Meanwhile,another 24 mice were randomized into 3 groups(drug treatment group,control group and normal group),which underwent gastrogavage at the previously determined optimal dose of HPQP prior to irradiation at 9.0 Gy γ-ray except the normal group.All the mice were sacrificed 9 days after radiation,and the effects of the drug on the hematopoietic function were evaluated by spleen colony method,bone marrow nucleated cell counts and observation of the bone marrow pathology.Results Hydroxypiperquin phosphate can promote the post-radiative restoration of peripheral leukocytes to a normal level(P0.01) in mice,decreased mortality(P0.01),and markedly increased colony-forming unit-spleen(CFU-S) and bone marrow nucleated cell counts(P0.01).Conclusion Hydroxypiperquin phosphate has protective effect on the radiation-induced damage in mice;mice
