Inhibition effect of Pinellia extract fraction on Ca Ski tumor-bearing nude mice in different drug-delivery way

Abstract
Objective: To observe the growth inhibition effect of Pinellia extract fraction(PE) on transplanted tumor bearing nude mice, and to explore the best drug-delivery way of PE. Methods: Thirty nude mice models of Ca Ski were established. When transplanted tumor had increased to 100mm3, the nude mice were randomly divided into 5 groups: control group, intragastric group, intraperitoneal group, subcutaneous group and cisplatin group. The diameter and volume were measured and calculated, and the growth curve of tumor was also delineated. Nude mice were sacrificed at the 22 nd day, the weight of tumor was measured and the inhibitory rate was calculated. Histomorphology of the tumor was observed by using HE staining. Immunohistochemistry was used to test the expression of Ki-67. Results: The weights of the tumor of intragastric group, intraperitoneal group and cisplatin group were significantly lower than that of control group and subcutaneous group(P0.05), and the tumor inhibition rate of intragastric group, intraperitoneal group and cisplatin group were 38.71%, 31.86% and 45.92% respectively. The expression of Ki-67 in tumor of intragastric group and intraperitoneal group was significantly lower than that of control group and intraperitoneal group(P0.05). Conclusion: PE could inhibit the growth Ca Ski transplanted tumor in nude mice, and the best drug-delivery way is intragastric administration and intraperitoneal injection. In considering of better compliance in animal with intragastric group and it's a common medicine administration way, so it might tend be chosen in future experiments. One of the anti-tumor mechanisms in vivo might be related to the down-regulation of Ki-67 expression.
