Abstract
Objective To investigate the role of oxidative stress on apoptosis in human endothelial cell line EA.hy926 induced by ethanol. Methods Cells were cultured and divided into different groups: control group,experimental group(0.6%ethanol,V/V) and NAC group(0.6% ethanol+ 10 mmol·L-1 NAC).Each group was incubated with corresponding drugs for 12 h.The inhibitory rate of cell proliferation was assessed by MTT method.The cell survival rate was evaluated by trypan blue exclusion test.The apoptosis rate and intracellular ROS were measured by flow cytometry assay.SOD activity and MDA level were detected by commercial kits. Results Compared with the control group,the experimental group resulted in a significant increase in the level of MDA from(46.5±3.8) nmol·mg-1 protein to(89.5±5.6) nmol·mg-1 one,the generation of ROS from(93.69±7.58) to(162.30±18.85) and the apoptosis rate from(0.49±0.16)% to(5.31±0.54)%,but a remarkable decrease in SOD activity from(40.8±2.9) U·mg-1 protein to(29.2±4.0) U·mg-1 one.The level of MDA,the generation of ROS and the apoptosis rate of NAC group were significantly lower than control group,while SOD activity improved noticeably. Conclusion Results indicate that oxidative stress may play an important role in ethanol-induced apoptosis.
