The Effect of VEGF on iNOS Protein Expression in HaCaT Cells and Its Relationship with the Cell Proliferation

Abstract

Objective To investigate the effect of VEGF on iNOS protein expression in HaCaT cells and the relationship with the cell proliferation. Methods The protein of iNOS in HaCaT cells was measured by Western blot; NO levels in HaCaT cellular supernatant were measured by Griess method; the effect of proliferation on HaCaT cells treated by VEGF and nitric oxide synthase inhibitor (L-NAME) were measured by MTT. Results Within the selected concentrations (5ng/mL,10ng/mL,25ng/mL,50 ng/mL) ,VEGF could promote the proliferation of HaCaT cells. The protein of iNOS and NO levels in HaCaT cells stimulated by VEGF was significantly increased. L-NAME could inhibit the proliferation of HaCaT cells stimulated by VEGF (10ng/mL) at 48h and in a dose-dependent manner. Conclusion VEGF could enhance iNOS protein expression in HaCaT cells. Meanwhile,the expression of iNOS protein might be closely correlated with the proliferation of HaCaT cells stimulated by VEGF.
