Induction of the Growth Inhibitation and Apoptosis in HL-60 Cell Line by 8-nitrochyrsin

Abstract

Objective:To investigate the effect of 8-nitrochyrsin(NOChR) on the growth and apoptosis of HL-60 cells cultured in vitro and its mechanism.Methods:8-nitrochyrsin on proliferation of HL-60 cells was detected by MTT assay.The trypan blue exclusion method was used to count the number of viable and dead cells,then the survival rate and growth curve were achieved.The apoptosis rate and cell cycle of HL-60 cells were determined by PI staining flow cytometry.Results:8-nitrochrysin inhibited proliferation of HL-60 cells in a concentration-dependent manner.The trypan blue exclusion method hint 8-nitrochyrsin posses the inhibitory effect of the growth of HL-60 cells.PI staining flow cytometry showed that cell cycle was arrested at G1 phase and the apoptosis of HL-60 cells was induced by NOChR in a concentration-dependent manner.Conclusions:NOChR posseses induction of apoptosis of HL-60 cells in vitro,and its mechanism may be associated with G1 phase cell cycle arrest.
