Expression of hTERT mRNA and correlation with telomerase activity and their clinical significance in colorectal cancer

Abstract

Objective:To investigate the expression of hTERT mRNA and correlation with telomerase activity and their clinical significance in colorectal cancer.Methods:In20specimens of human colorectal cancer and adjacent tissues,hTERT gene expression was detected by RT-PCR assays of hTERT mRNA,and telomerase activity was measured using TRAP-ELISA method.Results:The positive rates of hTERT mRNA expression and telomerase activity in colorectal cancer tissues were85%(17/20)and90%(18/20)respectively,and significantly higher than those in adjacent cancer tissues5%(1/20)(P0.0001).A significant correlation of telomerase activity level with differentiation of malignance and lymph node metastasis(P0.05)was observed,but not with tumor size,invasive depth,age or sex(P0.05).Concluˉsion:Telomerase activation may play an important role in the carcinogenesis and progression in human colorectal cancer.There was a strong positive correlation between hTERT mRNA exˉpression and telomerase activity.Both hTERT mRNA expression and telomerase activity can be used as diagnostic markers but the former is more simple and convenient than the latter.
