Study on culture of human embryo blastocyst and its related factors

Objectives To investigate the culture of human embryo blastocyst in vitro and its related factors. Methods The 54 IVF surplus embryos of day 3 from 18 volunteers were transferred to different mediums (IVC-Three medium,blastocyst medium or HTF medium) containing 10%SSS for advanced culture.The growth of embryos was observed until the embryo degeneration or 7 days after fertilization.Results The total blastulation rates of 54 embryos were 29.6%.The positive correlation between the cell number of day 3 embryo and blastulation rate was significant(r=0.99,P0.01).And the higher blastulation rate was positively related to embryo quality score (34.9% for 1～2.5 degree versus 9.1% for 3～4degree).The blastulation rate of IVC-Three medium was significantly higher than that of blastocyst medium and HTF(χ21=5.77,χ22=3.95,P0.05).Conclusion Advanced culture of surplus embryo will reach some blastulation.The cell number of day 3 embryo and quality score have some indication to blastulation.IVC-Three blastocyst medium may have potential clinical use for embryo blastocyst culture.
