Abstract

Objective:To establish a method for determination of mannitol in rabbit plasma. Methods:The plasma samples was deproteinized with acetate trichloride, mannitol contained in supernatant was oxidated with sodium periodate deproteinized and further reacted with Nash reagent to form the colour product which can be detected at a wavelength of 405 nm. Results:The linear range of calibration curve was 0.188-6.00 mg·mL -1, r=0.999 7(n=6). Corresponding to the drug plasma concentration. The average rate of recovery was 98.30%. RSD was 1.89%(n=3). Conclusion:The method is rapid, simple and can be used for determination of plasma concentration of mannitol and its clinical pharmacokinetic study.
