Simultaneous determination of geniposide,baicalin and berberine hydrochloride in Daochi Pills by multiple wavelength RP-HPLC

Abstract

AIM To establish a method for simultaneously determining geniposide,baicalin and berberine hydrochloride in Daochi Pills(Coptidis Rhizoma,Gardeniae Fructus,Scutellariae Radix,Rhei Radix et Rhizoma,Forsythiae Fructus,Akebiae Caulis,Scrophulariae Radix,Trichosanthis Radix,Paeoniae Radix rubra,Talci Pulvis).METHODS A multiple wavelength RP-HPLC method was developed.The analysis was performed on the column of Zorbax Eclipse XDB-C18(150 mm × 4.6 mm,5 μm) with gradient elution by using acetonitrile(A) and 0.2% phosphoric acid solution in the order of A: 15%-20%(0.00-10.00 min),A: 20%-25%(10.00-15.00 min),A: 25%-35%(15.00-25.00 min).The flow rate was 1.0 mL/min and the column temperature was maintained at room temperature.Geniposide,baicalin and berberine hydrochloride were detected at 240 nm,278 nm and 340 nm,respectively.RESULTS The calibration curves of geniposide,baicalin and berberine hydrochloride showed good linearity in the ranges of 0.037 5-0.337 5 μg(r=0.999 7),0.036 8-0.736 μg(r=0.999 9) and 0.039-0.351 μg(r=0.999 9),respectively.The average recoveries of geniposide,baicalin and berberine hydrochloride were 102.95%(RSD of 2.63%),102.83%(RSD of 1.90%) and 98.61%(RSD of 2.98%),respectively.CONCLUSION The method is simple,accurate and reproducible for simultaneous determination of geniposide,baicalin and berberine hydrochloride in Daochi Pills.
