Abstract
Objective To study the direct effects of gradient concentration interferon α(IFN α) and arsenic trioxide (As 2O 3 ) on EB virus infected Burkitt lymphoma cells line Raji and EBV negative cell line Daudi.Methods We have tested the effects of the combination of IFN α and As 2O 3 on cell proliferation,cell cycle phase distribution,and apoptosis in Raji and Daudi cell line by MTT,flow cytometry,Annexin V ,TUNEL and electron microscope.Results Low dose IFN α did not any significant effects on growths of Raji and Daudi cells,high dose IFN α(10 000 U/mL) have significant anti proliferative effects with dependence of time and dosage on both Raji and Daudi cells.There was a dose dependent accumulation of these cell in the G 0/G 1 phase of the cell cycles 24-48 hour from the time of exposure to IFN α,S phase of the cell cycles decline,increasing degree of apoptosis followed between 72 and 96 hours,IFN α and arsenic trioxide synergize to induce cell cycle arrest and apoptosis.EBV infected Raji cell more sensitive to IFN α/As 2O 3 than Daudi cell.Conclusions A high synergnistic effect between IFN α and As 2O 3 was observed in Raji and Daudi cell,with a dramatic inhibition of cell proliferation,G 0/G 1 arrest and induction of apoptosis.
