Abstract
The effect that different culture factors exerted on callus induction and differentiation of Amorphophallus were researched. The results showed that when culture conditions were MS + 2.0 mg / L NAA + 1.0 mg / L 6-BA or MS + 2.0 mg / L 2,4-D +0.5 mg / L KT or MS + 2.0 mg / L 2,4-D + 1.0 mg / L KT, optimal callus induction rate were obtained. Four different explants had significant influence on callus induction and the rate of induction was coleoptile corm learroot. Moreover, AC was able to enhance callus induction rate. Besides, the effect of buds differentiation was optimum when the culture condition was MS+2.0mg / L TDZ+0.1 mg / L NAA, with the rate and the ratio of differentiation being 96.15% and 9.52.
