Abstract
Objective To observe rat osteoclast apoptosis during cell culture in vitro.Methods Rat osteoclasts were cultured through mechanic separated technology and identified through morphology,Tartrate-resistant acid phosphatase staining and osteoclastic resorption test.The present experiment was divided into four groups,including 12h group,24h group,48h group and 72h group.Osteoclasts in respective culture time were counted after TRAP staining.Results Osteoclasts of large size were positive to TRAP staining,and could form resorptive lacunae.TRAP positive osteoclast amount took on a decreasing tendency after cell culture time was extended.Conclusion Rat osteoclast amount was decreased because of apoptosis during cell culture.
