A Comparison of LPS and TNF-α Induced ICAM-1 Expression of Vascular Endothelial Cells

Abstract

Objectve and methods To compare the difference of expression of intercellular adhesion molecule_1(ICAM_1) on human umbilical vein endothelial cell(HUVEC) induced by lipopolysaccharide(LPS) and tumor necrosis factor_α(TNF-α) by cellular immune fluorescent staining and confocal microscope. Results Comparing with basic expression of ICAM-1 on HUVEC, the ICAM-1 expression was enhanced significantly after 8 h to 24h of LPS stimulation, then descended at 36h obviously. the ICAM-1 expression induced by TNF-α was also increased significantly after 8h to 24h, and still maintained high level at 36h. Dose_dependent relationship existed between ICAM-1 expression and LPS or TNF-α. Conclusion The expression of ICAM-1 on HUVEC could be upregulated by LPS and TNF-α significantly, but tolerated to LPS chronical stimulation.
