EFFECTS OF XIONGSHAO CAPSULE MEDICATED SERUM ON EXPRESSION OF VEGF,Ang-2 mRNA IN HUMAN UMBILICAL VEIN ENDOTHELIAL CELLS

Objective:To explore the molecular mechanism of pro-angiogenic effect of xiongshao capsule by observing the effects of xiongshao capsule medicated serum on vascular endothelial growth factor (VEGF ) and angiopoietin-2(Ang-2 )mRNA expression in human umbilical vein endothelial cells (HUVEC).Methods: Different doses (high,middle and low) xiongshao capsule medicated serum were added to human umbilical vein endothelial cells in vitro,the morphologic changes of HUVEC was observed by phase contrast microscope,the proliferation of HUVEC was determined by MTT assay,VEGF and Ang-2 mRNA expression were detected by reverse transcriptase-polymerase chain reaction(RT-PCR).Results: Three doses xiongshao capsule medicated serum could promote proliferation of HUVEC,the high dose could obviously increase the VEGF mRNA expression,the effects of middle and low doses weren't significant. The high and middle doses could inhibit Ang-2 mRNA expression,the effect of low dose wasn't significant.Conclusion: The pro-angiogenic effect of xiongshao capsule was related with promoting VEGF mRNA and inhibiting Ang-2 mRNA expression.
