Abstract

Objective To observe the dynamic changes of Micro-RNA 200(miR-200s) Family in rat liver during the development of chronic liver injury induced liver fibrosis.Methods Forty male Wistar rats weighing 180-220 g were divided into five groups(control group and four model groups).Model groups were induced by hypodermic injection of CCl4 for 2,4,6 and 8 weeks.The liver index and the content of ALT and AST in serum were analyzed,and the liver fibrosis were observed by microscope.The expression of miR-200a,miR-200b,miR-200c,miR-141 and miR-429 mRNA were determined by using Real-time PCR respectively.Results The liver index,ALT and AST in four model groups were obviously higher than normal control group.The apparent liver fibrosis was observed in 8 weeks model group.The expression of miR-200a,b,c,141 and 429 mRNA in rat liver of 8 weeks model groups were obviously higher than that in control.(P0.05).Conclusion In the process of liver fibrosis induced by CCl4,the obvious change of miR-200s reveals that the miR-200s may play an important role in the chronic liver injury and liver fibrosis.
