Abstract
Objective To study the anti-bladder tumor effect of Ag85A DNA vaccine in tumor-bearing mice.Methods Agroupof5~6weeksT739micewererandomlydivided into four group:pcDNA3.1-Ag85A group,pcDNA3.1 group,NS group and BCG group.BTT739 cells(1×106)in their vigorous growth were inoculated subcutaneously into left posterior limb of each mouse in every groups.0.1 mL pcDNA3.1-Ag85A,pcDNA3.1,NS or BCG was inoculated into left posterior limb muscle at the immuned time that is the seventh day,fourteenth day and twenty-first day after tumor bearing.At the twenty-eighth day,the mice were sacrificed.The tumors were removed to measure the tumor weight and the tumor inhibition rate was calculated.Spleen was removed aseptically.The percentages of CD4+T cell and CD8+T cell were measured by flow cytometry.In addition,IFN-γ in sera of the tumor-bearing mice were assayed with indirect enzyme-linked immuno-absordent assay(ELISA).Results The results of tumor inhibition rate and the percentage of CD4+T cell,CD8+T cell and the relative value of CD4+/CD8+ showed that there wasn't difference among the Ag85A DNA vaccine group,pcDNA3.1 group and NS group.The production of IFN-γ showed that there was difference among the Ag85A DNA vaccine group,pcDNA3.1 group and NS it couldn't come up to BCG level.While the pcDNA3.1 group and NS group have no significant changes.Conclusion Intramuscular injection of Ag85A DNA vaccine may not induce the effective immunological effect in tumor-bearing.
