Abstract

Objective:To compare the expression of NCAM subtype between the transfection to different cell lines.Methods:FuGENE HD Transfection Reagent and LlipofactaneTM2000 were used to transfect the SH-SY5Y cells or HEK293 cells with plasmids which were respectively encoding NCAM180,NCAM140 or NCAM120,and the efficiency was detected by immunofluorescence method and Western blotting.Results:In the research,after transfecting the plasmids into the SH-SY5Y cells by LlipofactaneTM2000 and FuGENE HD Transfection Reagen,we can't detect the high expression of NCAM120,-140,or-180 by the immunofluorescence method or Western blotting.The result of western blotting show that,after transfecting the plasmids into the HEK293 cells by FuGENE HD Transfection Reagen,the NCAM180,-140 and-120 could be highly expressed respectively.Conclusion:By the transfection,the SH-SY5Y cell line couldn't highly express NCAM140,-120 and-180,while the HEK293 cell line can highly express NCAM120,-140,and-180.
