Abstract
Objective To investigate the effect of sodium ferulate(SF)on the expression of α-smooth muscle actin(α-SMA),connective tissue growth factor(CTGF) and the extracellular matrix(ECM) in renal fibroblasts stimulated by TGF-β1.Methods Normal rat renal fibroblasts(NRK-49F) were cultured in vitro.The effect of SF on the TGF-β1 induced cell vitality was observed with MTT.The expressions of α-smooth muscle actin(α-SMA) and connective tissue growth factor(CTGF) were measured with real-time quantitative PCR and immunocytochemistry.The synthesis of collagen type I(Col I) and fibronectin(FN) was determined with ELISA.Results TGF-β1 markedly increased the cell vitality,the expression of α-SMA and CTGF and the synthesis of Col I and FN in NRK-49F,which was partly attenuated by SF.Conclusion SF can inhibit renal fibrosis induced by TGF-β1.
