Abstract
Objective To study the effect of daizongfang on HepG2 intracellular lipid accumulation induced by oleic acid. Methods Oleic acid was used to induce liver cell model of HepG2 intracellular lipid accumulation. MTT experiment was adopted to determine the impacts on HepG2 cell viability with oleic acid of different concentration or daizongfang. DODIPY493 /503 and DAPI double fluorescent staining was adopted for the morphological observation of intracellular lipid accumulation. MATLAB software was used for the mathematical treatment of fluorescent pictures and to measure the content of intracellular lipid. Results Oleic acid 100- 800μm did not brought significant impacts on HepG2 cell survival rate( P 0. 05). The intracellular lipid accumulation in HepG2 induced by oleic acid was positively relevant with the dose. Daizongfang of certain dose( 1 mg / L ~ 20 mg / L) did not impact significantly HepG2 survival rate( P 0. 05) and significantly reduced liver cell lipid content induced by oleic acid. Daizongfang 10 mg / L worked in 4 h and the effect was stable in 96 h. Daizongfang 40 mg / L reduced HepG2 survival rate( P 0. 05) and had no significant impact on intracellular lipid content( P 0. 05). Conclusion Daizongfang inhibits HepG2 intracellular lipid accumulation induced by oleic acid.
