Alteration of IFNAR and IL-17 in CHB patients after IFN-α treatment

Abstract

【Objective】To study the alteration of IFNAR and IL-17 in chronic hepatitis B(CHB) patients after IFN-α treatment. 【Methods】26 cases of CHB patients were enrolled in this study. The patients were divided into low HBV-DNA copy group(n =12),high HBV-DNA copy group(n =14),IFN-α treatment response group(n =13),and IFN-α treatment non-responsegroup(n =12). Another 10 no-HBV person were used as control group. The mRNA levels of IFNAR were measured by Real-time PCR analysis. The serum IL-17 was measured by ELISA analysis. 【Results】The mRNA expression of IFNAR in CHB patients was(0.216±0.107),which was significantly higher than that of(0.114±0.082) in control group(P 0.05). The IL-17 expression in CHB patients was(427.3 ±126.4) pg/mL,which was significantly higher than that of(304.5±114.8) pg/mL in control group(P 0.05). The mRNA expression of IFNAR in high HBV-DNA copy group was(0.254±0.116),which was significantly higher than that of(0.172±0.093) in low HBV-DNA copy group(P 0.05). The IL-17 expression in high HBV-DNA copy group was(481.6±142.5) pg/mL,which was significantly higher than that of(372.7±123.5) pg/mL in low HBV-DNA copy group(P 0.05). IF-NAR expression in CHB patients was significantly positively correlated with IL-17 expression(r =0.482,P 0.05). The mRNA expression of IFNAR in response group was(0.324±0.122),which was significantly higher than that of(0.231±0.109) in non-response group(P 0.05). The IL-17 expression inresponse group was(324.6±114.2) pg/mL,which was significantly lower than that of(414.7±127.8) pg/mL in non-response group(P 0.05). IFNAR expression was significantly negatively correlated with IL-17 expression after IFN-α treatment(r =-0.305,P 0.05).【Conclusion】The liver tissue expression of IFNAR and blood expression of IL-17 were increased in CHB patients. After interferon treatment,the IFNAR expression was upregulated,and IL-17 expression was decreased in effective group. Detection of the two indicators maybe contribute to predict the clinical efficacy of interferon treatment of CHB
