Abstract
Objective To study the signal-transduction mechanism of ventricular remodeling after myocarclial infarction in rats and the effect of perindopril. Methods After the model of myocardial infarction was established by ligating the left anterior descending coronary of rats, the survivals were randomly divided into two groups: group A (myocardial infarction group, n=14) and group B (perindopril group, n=14). Meanwhile, the other 14 shame operation rats were as control (group C, n=14). Each group was treated with direct gastric gavage from the second day, and rats of group B was treated with perindopril, group A and C with pure water. Six weeks later, the AngⅡcontent was detected through radioimmunoassay, the ERK1/2 content was detected with immunohistochemical method and the expression of c-fos mRNA through real-time fluorescent quantitative PCR. Results Compared with group C, there was a significant increase of AngⅡand ERK1/2 content, expression of c-fos mRNA and left ventricular weight index in group A and B (P 0.05). Compared with group A and B, the AngⅡand ERK1/2 content, the level of c-fos mRNA expression and left ventricular weight index were lower in group B than group A(P0.05). Conclusion After myocardial infarction, the AngⅡand ERK1/2 content and the expression of c-fos mRNA in myocardium and left ventricular weight index is increased distinctly. Perindopril can remarkably reduce the expression of c-fos mRNA and the content of AngⅡand ERK1/2 in myocardium.
