Effect of BDNF on the expression of NF-κB in acute spinal cord injury rat

Abstract

Objective To study the expression of nuclear factor-kappa B(NF-κB) in acute spinal cord injury rat after BDNF was injected.Methods The spinal cord injury was induced with Nystrom way.The healthy adult Wistar rats(72) were randomly divided into normal control group,spinal cord injury group,BDNF group on average.The expression of NF-κB was observed by immunochemistry and western blot methods in all groups.Results The mean optic density(A) of NF-κB positive product increased significantly in acute spinal cord injury group and BDNF group than in normal control by means of immunochemistry(P 0.01);but decreased in the BDNF group than in the acute spinal cord injury group(P 0.01).The ratios between integrated density value(IDV) for NF-κB protein band and IDV for β-actin protein band increased significantly in the spinal cord injury group and BDNF group than in normal control by means of Western blot(P0.01);but decreased in the BDNF group than in the spinal cord injury group(P0.01).Conclusion One of the mechanisms for BDNF participate the pathogenesis of acute spinal cord injury might be upregulate the expression of NF- κB.
